General Information of the m6A Target Gene (ID: M6ATAR00652)
Target Name CCR4-NOT transcription complex subunit 7 (CNOT7)
Synonyms
BTG1-binding factor 1; CCR4-associated factor 1; CAF-1; Caf1a

Gene Name CNOT7
Chromosomal Location 8p22
Family CAF1 family
Function
Has 3'-5' poly(A) exoribonuclease activity for synthetic poly(A) RNA substrate. Its function seems to be partially redundant with that of CNOT8. Catalytic component of the CCR4-NOT complex which is one of the major cellular mRNA deadenylases and is linked to various cellular processes including bulk mRNA degradation, miRNA-mediated repression, translational repression during translational initiation and general transcription regulation. During miRNA-mediated repression the complex seems also to act as translational repressor during translational initiation. Additional complex functions may be a consequence of its influence on mRNA expression. Associates with members of the BTG family such as TOB1 and BTG2 and is required for their anti-proliferative activity.

YTH domain-containing family protein 1 (YTHDF1) [READER]
In total 1 item(s) under this regulator
Experiment 1 Reporting the m6A Methylation Regulator of This Target Gene [1]
Response Summary YTHDF1 could recognize the m6A sites of CCR4-NOT transcription complex subunit 7 (CNOT7) and promote its expression in an m6A manner. Inhibition of YTHDF1 could suppress the proliferation, migration and invasion of the OS cells.
Target Regulation Up regulation
Responsed Disease Osteosarcoma ICD-11: 2B51
Cell Process Cell proliferation
Cell migration
Cell invasion
In-vitro Model U2OS Osteosarcoma Homo sapiens CVCL_0042
SaOS-2 Osteosarcoma Homo sapiens CVCL_0548
MG-63 Osteosarcoma Homo sapiens CVCL_0426
HOS Osteosarcoma Homo sapiens CVCL_0312
hFOB 1.19 Normal Homo sapiens CVCL_3708
In-vivo Model The mice were divided into two groups with three mice in each group and their flank was subcutaneously injected with 1 × 107 OS cells. 28 days following subcutaneous injection, the mice were sacrificed through carbon dioxide euthanasia (30%/min) to obtain tumor weight and volume measurements.
Osteosarcoma [ICD-11: 2B51]

Experiment 1 Reporting the m6A-centered Disease Response [1]
Response Summary YTHDF1 could recognize the m6A sites of CCR4-NOT transcription complex subunit 7 (CNOT7) and promote its expression in an m6A manner. Inhibition of YTHDF1 could suppress the proliferation, migration and invasion of the OS cells.
