| In-vivo Model | Male nude mice (age: 4-6 weeks, weight: 22-22 g) provided by the Southern Medical University, were housed in specific pathogen environment at 22-24°C and 50-60% humility under a 12-h light/dark cycle, with ad libitum access to food and water. The nude mice were classified into 3 groups with 18 mice per group: the control group, the sh-NC group, and the sh-METTL3 group. Well-grown ECA109 cells, sh-METTL3-transfected ECA109 cells, sh-NC-transfected ECA109 cells were re-suspended in phosphate buffer saline (PBS) solution and then subcutaneously injected (1 × 107cells/mouse) into the right abdomen (N = 6) or injected into mice via the caudal veins (N = 6). For the subcutaneously injected mice, the tumor volume was evaluated every 7 days according to the following formula: V = ab2/2 (a: the longest diameter of the tumor; b: the shortest diameter of the tumor). After 30 days, the subcutaneously injected mice were euthanatized using an intraperitoneal injection of 1% pentobarbital (200 mg/kg). | ||||
